Molecular characterization and phylogenetic of newcastle disease virus strains, isolated from rural areas of Southern Ecuador

Newcastle disease virus is a highly contagious avian paramyxovirus and the main devastating poultry agent, causing high economic losses in the poultry industry around the world. In the present study, we characterized molecularly 8 partial gene sequences of the fusion protein and 3 complete sequences of the haemagglutinin-neuraminidase from isolated virus present in native poultry unvaccinated in the Southern Ecuador in 2014. These sequences were compared with other sequences of virus, representing different genotypes and sub-genotypes of virus from different regions of the world. All viruses showed the amino acid sequence 112GRQGRL117 in C-terminus extension of the F2 protein, and showed Leucine at the N-terminus extension of the F1 protein. Those sequences allowed us to determine that these viruses belonged to a type of lentogenic virus. Phylogenetic analysis showed that most of the Newcastle virus isolated in this region of Ecuador, were closely related to strains of genotype II Class II, consistent with the sequence lentogenic live-virus vaccine, La Sota and B1. Molecular characterization and phylogenetic study of the F and HN genes could reliably be used as a tool for monitoring and early detection of Newcastle disease virus, as well as to predict the different pathotypes of virus isolates.Metrics
